El nostre grup es centra en la oncoproteïna pleiotròpica i ubiqua Myc, la desregulació de la qual està implicada en gairebé tots els càncers humans. Els desafiaments tècnics d’apuntar a factors de transcripció nuclear com Myc, i la preocupació pels possibles efectes secundaris, havien exclòs fins fa poc qualsevol validació preclínica de la inhibició de Myc com a possible estratègia terapèutica.

En els últims anys, hem demostrat en diversos models murins que la inhibició de Myc té un profund impacte terapèutic en diversos tipus de tumors, amb efectes secundaris molt lleus i reversibles en el teixit normal.

Estimulats pels nostres resultats en ratolins, ara estem interessats a desenvolupar opcions farmacològiques viables i no tòxiques destinades a Myc a la clínica. Per a això, hem creat una empresa derivada, Peptomyc S.L., per al desenvolupament de pèptids inhibidors de Myc per al tractament del càncer. El nostre laboratori, en col·laboració amb Peptomyc, està validant actualment el nostre nou enfocament contra càncers notòriament difícils de tractar que són resistents als tractaments de referència i que necessiten urgentment noves vies terapèutiques (és a dir, càncer de pulmó no microcític provocat per KRAS, glioblastoma i càncer de mama triple negatiu amb metàstasi).

El nostre grup ha seguit contribuint a la innovació científica amb publicacions en revistes de prestigi internacional. Destaquem un article, publicat el 2019, que descriu els estudis clau per al progrés del nostre inhibidor de Myc, la miniproteína Omomyc, cap a l’ús clínic (Beaulieu et al. Intrinsic cell-Penetrating activity propels Omomyc from proof of concept to viable anti Myc therapy. Sci Trans Med. 2019). Aquesta publicació va ser molt ben rebuda per la comunitat científica i destacada a Nature Reviews Cancer i Nature Reviews Drug Discovery com un possible fita fonamental cap a l’aplicació clínica del nostre inhibidor Myc primer en la seva classe.

OBJECTIUS ESTRATÈGICS

  • Validació de nous pèptids penetrants de cèl·lules basats en Omomyc per al tractament del càncer.
  • Validació preclínica de noves teràpies anti-Myc en el càncer de mama, cervell, pulmó, neuroblastoma, melanoma, càncer colorectal i mieloma múltiple.
  • Definir el paper de Myc en la promoció de l’evasió immunitària de el càncer.
  • Dilucidar el paper de la xarxa Myc en els tumors de l’estroma gastrointestinal (tegi) amb defectes màxims, el càncer de pulmó microcític (CPMC) i els feocromocitomes (FCC).

ASPECTES DESTACATS

  • La publicació de Beaulieu et al. a Science Translational Medicine, que demostra que Omomyc, el pèptid inhibidor de Myc, pot penetrar en les cèl·lules canceroses i actuar contra els tumors in vivo, demostrant el seu potencial per al desenvolupament clínic, ha generat una gran cobertura mediàtica.
  • Laura Soucek i Jonathan Whitfield van ser editors convidats d’una edició especial de Current Opinion in Pharmacology sobre pèptids en el càncer.
  • Nou finançament addicional concedit, en particular de La Marató i FIS (ISCIII).
